Radiopharmaceuticals
1: TECHNETIUM LABELLED DENATURED RED BLOOD CELLS.
(Merseyside And Cheshire Radiopharmacy Services).
Materials required.
Technetium generator eluate 1 x 10ml syringe
1 x Stannous Chloride vial (Amersham stannous agent N106) 1 x 5ml syringe
1 x 4ml ACD vial 1 x 1ml syringe
3 x 10ml sodium chloride 0.9% 3 x 19g needle
1 x 0.22 micron filter 2 x 21g needle
2 x 60ml tubes
Shaker/ Water bath set at 49.5°C.
Method.
1. Start up the heater/shaker bath in order for it to reach the correct temperature.
2. Make up the Stannous kit in 6ml sodium chloride 0.9%, 2ml of which is to be dispensed to a patient.
3. 20 minutes post injection of Stannous, 16ml of patients blood is taken and placed into the vial containing 4ml ACD (acid citrate dextrose) solution.
4. The sample is then centrifuged at 2000 rpm for 10 minutes, and the plasma (supernatant) removed.
5. Add 200MBq of technetium to the cells agitating gently. Incubate for 15 minutes at 49.5°C in the shaker/water bath.
6. Add 10 ml sodium chloride and centrifuge at 2000 rpm for 10 minutes then remove the supernatant.
7. Repeat 6. above.
8. Re-disperse the red cells with sodium chloride 0.9% to a total volume of 15 ml.
9. Measure the activity and dispense the required amount.
