| Materials required. | ||
| Technetium generator eluate | 1 x 10ml syringe | |
| 1 x Stannous Chloride vial (Amersham stannous agent N106) | 1 x 5ml syringe | |
| 1 x 4ml ACD vial | 1 x 1ml syringe | |
| 3 x 10ml sodium chloride 0.9% | 3 x 19g needle | |
| 1 x 0.22 micron filter | 2 x 21g needle | |
| 2 x 60ml tubes | ||
| Shaker/ Water bath set at 49.5°C. | ||
| Method. | ||
| 1. | Start up the heater/shaker bath in order for it to reach the correct temperature. | |
| 2. | Make up the Stannous kit in 6ml sodium chloride 0.9%, 2ml of which is to be dispensed to a patient. | |
| 3. | 20 minutes post injection of Stannous, 16ml of patients blood is taken and placed into the vial containing 4ml ACD (acid citrate dextrose) solution. | |
| 4. | The sample is then centrifuged at 2000 rpm for 10 minutes, and the plasma (supernatant) removed. | |
| 5. | Add 200MBq of technetium to the cells agitating gently. Incubate for 15 minutes at 49.5°C in the shaker/water bath. | |
| 6. | Add 10 ml sodium chloride and centrifuge at 2000 rpm for 10 minutes then remove the supernatant. | |
| 7. | Repeat 6. above. | |
| 8. | Re-disperse the red cells with sodium chloride 0.9% to a total volume of 15 ml. | |
| 9. | Measure the activity and dispense the required amount. | |
