6: METHOD FOR CONCENTRATING 99mTc GENERATOR ELUATE (For 99mTc-technegas preparation)
(Merseyside And Cheshire Radiopharmacy Services).
Materials Required
1 x Onguard -AG column (Dionex Ltd)
1 X BondElut SAX column (Jones Chromatography)
1 x BondElut adapter (Jones Chromatography)
Water for injection
Ethanol
Sodium Chloride 5% solution
Nitrogen filled vial
2 x 21g (green) needles
1 x 23g (blue) needle
1 x 1ml syringe
3 x 5ml syringes
1ml and 5ml syringe shields
lead pot
99mTc generator eluate.
Method
1. Wash Onguard-AG column with 5ml water for injections.
2. Wash BondElut SAX column with 3ml ethanol, followed by 3ml water for injections.
3. Connect Onguard-AG column to BondElut-SAX column using a BondElut adapter.
(MAKE SURE THE CONNECTIONS ARE TIGHT)
4. Place the lower, BondElut SAX column in a suitable lead shield.
5. Pass 5ml of 99mTcO4- generator eluate (in a shielded syringe) through the column system, followed by 2ml water for injections. Pass slowly (5ml/min).
6. Then pass 5ml of air through the system. Use the syringe formerly holding water for injections.
7. Run effluents to waste.
8. Disconnect Onguard-AG column and discard.
9. Fit BondElut-SAX column with green needle (21g). Fit a sterile nitrogen filled vial with a bleed needle (23g) and place in a lead shield.
10. Pierce septum of N2 vial with BondElut SAX column/needle.
11. Elute column with 1ml sodium chloride 5%, collecting eluate in vial.
12. Pass through column 2ml air to ensure maximum extraction of activity.
13. Calibrate product, label and examine with TLC, if necessary. (Si-gel, n-butanol/acetic acid/water 3:2:3)
